ZXH-8-004 is a selective and potent PROTAC degrader of the envelope (E) protein of dengue virus (DENV), demonstrates potent antiviral activity.
ZXH-8-004 is cell permeable and able to engage the CRL4CRBN E3 ligase.
ZXH-8-004 has excellent selectivity for E with the only other significantly degraded proteins being the well-known 'neosubstrates' derived from the glutarimide recruiter (ZFP91 and GSPT1).
ZXH-8-004 reduces intracellular E starting at a 1.25 μM dose in DENV2-infected Huh7.5 cells.
ZXH-8-004 (2.5, 5 uM) demonstrate markedly increased antiviral efficacy across various serotypes compared to parental inhibitor CVM-2-12-2
