XY-07-35 is a potent, selective HDAC6 proteolysis targeting chimera (PROTAC) degrader with IC50 of 48.5 nM.
Treatment of cells with XY-07-35 leads to recruitment of the E3 ligase cereblon (CRBN) to HDAC6, ubiquitination and subsequent proteasomal degradation of HDAC6.
XY-07-35 displays 10 times selectivity over HDAC8 (IC50=0.621 μM) or other HDACs 168 (IC50=1.04-5.75 μM).
XY-07-35 (1 uM) efficiently degraded HDAC6 in PBMCs, does not cause HDAC6 degradation in both non-differentiated and PMA differentiated THP-1 cells.
