WIZ degrader 10 is a potent, and selective molecular glue degrader of the transcription factor WIZ with DC50 of 3 nM and Dmax of 86%.
WIZ degrader 10 induces endogenous WIZ degradation in primary human erythroblasts with DC50 of 2 nM, a maximum depth of 84%, demonstrates a ∼ 9x increase in potency over dWIZ-2.
WIZ degrader 10 increases potency of WIZ degradation translated to an increase in in vitro HbF induction compared with dWIZ-2. WIZ degrader 10 (0.01-1 mg/kg) in duces dose-dependent degradation of WIZ and HbF induction in a humanized NBSGW mouse model.
