WEHI-991 (W-991) is a potent small molecule inhibitor of PfSTART1 with antiparasitic activity EC50 of 7 nM for Pf 3D7, binds to PfSTART1(WT) with KD of 10 nM, inhibits ring-stage development of Plasmodium falciparum.
The PfSTART1 mutations confer resistance to, bu still reteins with sub-micromolar potency against the mutant parasites with an EC50 below 700 nM.
W-991 (WEHI-991) blocks Plasmodium falciparum ring development but the effect is reversible.
W-991 (WEHI-991) blocks differentiation into ring-stage parasites by preventing parasitophorous vacuole membrane (PVM) expansion.
W-991 (WEHI-991) does not prevent sporozoite invasion but blocks parasite transmission to mosquitoes.
