UNC6852 (UNC-6852) is a chemical degrader of PRC2, contains an EED226-derived ligand and a ligand for VHL, induces proteasomal degradation of PRC2 components, EED (DC50=0.61 uM), EZH2WT/EZH2Y641N (DC50=0.67 uM) and SUZ12.
UNC6852 selectively degrades PRC2 via the E3 ligase ubiquitylation pathway.
UNC6852 reduces H3K27me3 levels and DLBCL cell proliferation.
UNC6852 potently degrades the core components of PRC2 and results in a concomitant loss of H3K27me3 in DLBCL cells with EZH2 gain-of-function mutations.
UNC6852 decreases cell proliferation in EZH2 mutant DLBCL cell lines.
