UNC10088 is a highly potent, second-generation NSD2 degrader with DC50 of 0.06 uM and Dmax of 97%, facilitates FBXO22-mediated degradation of NSD2 in NSD2-mutant ALL.
UNC10088 is not only more potent, but also degrades NSD2 more rapidly compared to UNC8153.
UNC10088 causes more NSD2 degradation as compared to UNC8732, potentially due to the more rapid conversion of UNC10088 to the active aldehyde.
UNC10088 recruits the SCFFBXO22 ubiquitin ligase complex for compound-mediated degradation of NSD2, binds and recruits FBXO22 via interactions with C326.
