TBOPP is a selective inhibitor of Rac-specific guanine nucleotide exchange factor DOCK1, inhibits DOCK1-mediated Rac activation with IC50 of 8.4 uM.
TBOPP binds to DOCK1 DHR-2 domain with Kd of 7.1 uM.
TBOPP dampens DOCK1-mediated invasion, macropinocytosis, and survival under the condition of glutamine deprivation without impairing the biological functions of DOCK2 and DOCK5 proteins.
TBOPP blocks cancer cell invasion and survival in vitro, suppresses cancer metastasis and growth in vivo in mice.
