SecinH3 is a selective inhibitor of small GEFs of the cytohesin family, blocks human, mouse and Drosophila cytohesins with IC50 of 2.4 to 5.6 uM, inhibits ARF guanine nucleotide-exchange factors such as ARNO and ARF6.
SecinH3 binds to the Sec7 domain of these small Arf-GEFs but does not bind Sec7 domains of larger Arf-GEFs.
small GEFs prevents cytohesin-dependent insulin signaling in HepG2 cells, produces hepatic insulin resistance in mice.
