SR-717 (SR717) is a non-nucleotide, small-molecule STING agonist, demonstrates broad interspecies and interallelic specificity with EC50 of 2.1 uM and 2.2 uM in ISG-THP1 (WT) and ISG-THP1 cGAS KO (cGAS KO) cell lines.
SR-717 functions as a direct cGAMP mimetic that induces the same "closed" conformation of STING.
SR-717 displays antitumor activity, promotes the activation of CD8+ T, natural killer, and dendritic cells in relevant tissues, and facilitates antigen cross-priming.
SR-717 induces the expression of clinically relevant targets, including PD-L1, in a STING-dependent manner.
