RDN8011 is a potent, selective first-in-class PROTAC degrader of KDM4 with DC50 of 37-50 nM for KDM4A/KDM4B/KDM4C in in KYSE-150 cells, while sparing KDM4D.
RDN8011 acts KDM4A degradation mediated by the ubiquitin-proteasome system (UPS).
RDN8011 (0-10 uM) upregulates H3K36 methylation and induces cell cycle arrest and apoptosis in KYSE-150 cells.
