PfPK6 inhibitor Compound 12 is a potent, selective type II kinase inhibitor of Plasmodium protein kinase 6 (PfPK6) with IC50 of 301 nM, displays nanomolar activity against P. falciparum asexual blood stages with EC50 of 0.1 uM (P. falciparum Dd2 strain).
Compound 12 is a fast-acting compound active throughout the asexual blood cycle.
Compound 12 exhibits good pharmacokinetics and is active in vivo.
Compound 12 is not primarily mediated via PfPK6 inhibition.
Compound 12 inhibited βH formation with a mean IC50 of 13 uM.
Compound 12 is active against Cambodian and Ugandan P. falciparum isolates.
