NP3-742 is a potent, selective NLRP3 inhibitor, inhibits IL-1β release in supernatant of nigericin stimulated THP-1 cells with IC50 of 6 nM.
NP3-742 binds to the NLRP3 NACHT domain. Inhibits IL-1β release from LPS/ATP stimulated human whole blood with IC50 of 47 nM.
NP3-742 shows complete inhibition of IL-1β in the acute mouse peritonitis model when administered at 3 mg/kg p.o.
