MS140 (XY028-140) is a potent and selective, CRBN-dependent CDK4/6-degrader (PROTAC) with IC50 of 0.38 nM and 0.28 nM against CDK4/cyclin D1 and CDK6/cyclin D1, suppresses Rb/E2F signaling and reduces CDK4 and CDK6 protein levels.
MS140 does not degrade CDK7 or CDK9.
MS140 is more effective than CDK4/6 inhibitors in CDK4/6i-S tumor cells.
