MMV693183 is a first-in-class acetyl-CoA synthetase (AcAS) inhibitor and potent antimalarial drug candidate (Asexual blood stage IC50=2.1-2.8 nM).
MMV693183 is active against both early and late asexual blood stages, but is not efficacious against liver stages.
MMV693183 specifically inhibits female gametocyte activation with an IC50 value of 12 nM, whereas male gamete formation is inhibited much less with an IC50 value of 1 µM.
MMV693183 inhibits P. falciparum growth in an AcAS-dependent manner.
MMV693183-derived antimetabolite, CoA-MMV693183 directly binds and inhibits acetyl-CoA synthetase (AcAS), potently blocks P. falciparum transmission to Anopheles mosquitoes.
