MAR-3.6.2 is a specific small molecule, allosteric inhibitor of small GTPase RAN (RAS-related nuclear protein), targets RAN a cryptic pocket and disrupts its on-state and nuclear export, decreases miR-126 biogenesis.
MAR-3.6.2 reduces RAN GTP binding in HUVEC ECS.
MAR-3.6.2 has no measurable impact on the GTP-binding activity of related small GTPases, including RAS and RAB5.
MAR-3.6.2 disrupted RAN interaction with RCC1 and trapped RAN-GDP in the nucleus, decreased nucleus-to-cytoplasm export of pre-miR-126 and mature miR-126 biogenesis.
MAR-3.6.2 demonstrated dose-dependent inhibition of cell proliferation and induction of apoptosis across multiple AML cell lines.
MAR-3.6.2 (25 mg/kg, IV, daily for 3 weeks) exhibited significant antileukemic activity in the MllPTD/WT/Flt3ITD/ITD AML mouse model.
