KNX-002 (KNX002) is a potent, specific inhibitor of Toxoplasma gondii class XIV myosin TgMyoA, inhibits TgMyoA ATPase activity with IC50 of 2.8 uM, shows little to no effect on any of the vertebrate myosins.
KNX-002 has little to no detectable effect on the actin-activated ATPase activity of bovine cardiac myosin, chicken gizzard smooth muscle myosin, rabbit fast skeletal muscle myosin, or bovine slow skeletal muscle myosin at 40 uM.
KNX-002 inhibits parasite motility and growth in culture in a dose-dependent manner (IC50=19.7 uM), with no detectable toxicity to human cells.
The mutation TgMyoA (T130A) renders the motor less sensitive to KNX-002, parasites expressing the T130A mutation shows reduced sensitivity to KNX-002 in both motility and growth assays.
