JC124 is a potent selective NLRP3 inflammasome inhibitor with IC50 of 3.25 uM for inhibition of IL-1β release by LPS/ATP-stimulated J774A.1 cells.
JC124 (50 mg/kg daily, i.p. injection) inhibits the NLRP3 signaling in TgCRND8 mice, decreases Aβ deposition, reduces β-cleavage of APP and Aβ production.
JC124 reduces microgliosis but increases astrocytosis in TgCRND8 mice,.
JC124 reduces oxidative stress in TgCRND8 mice after long-term treatment, improves the level of synaptic marker.
