ATI-2307 (T-2307) is a pentamidine-like compound with antifungal activity, selectively targets fungal mitochondria via a fungal-specific transporter that is not present in mammalian cells.
ATI-2307 shows a broad spectrum of activity against many fungal pathogens including Candida spp, Aspergillus spp., and C. neoformans.
ATI-2307 shows IC50 of 0.01-0.06 ug/mL against C. neoformans clinical isolates from Uganda.
ATI-2307 demonstrates significant fungicidal activity at dosages between 1 and 2 mg/kg/d, and is superior to fluconazole and similar to amphotericin B treatment in rabbit model cryptococcal meningoencephalitis (CM).
ATI-2307 disrupts the mitochondrial membrane potential and inhibits the respiratory chain complexes of fungi-it thus represents a new mechanism for direct antifungal action.
