ASP0965 is potent, orally active and brain-penetrable NLRP3 inflammasome inhibitor with IC50 of 8.4 nM (in vitro IL-1β inhibitory activity).
ASP0965 also shows potent inhibition of ASC speck formation in THP-1 cells stimulated by LPS/ATP with IC50 of 5.7 nM.
ASP0965 significantly inhibits the production of IL-1β at 0.3, 1, and 3 mg/kg doses in rat cerebrospinal fluid (CSF).
ASP0965 shows a dose-dependent inhibition on α-synuclein-dependent IL-1β production in THP-1 cells with IC50 of 6.3 nM.
ASP0965 (10-mg/kg, qd) improves motor function in α-synuclein injection model.
