AN3661 is a specific small molecule inhibitor of Toxoplasma gondii CPSF3 (TgCPSF3), demonstrates potent activity against Toxoplasma gondii in vitro with IC50 of 0.9 uM.
The three mutated residues associated with AN3661 resistance (Y328C, Y483N and E545K) clustered to one side of the catalytic site of TgCPSF3.
AN3661 at 20 mg/kg orally for 7 days protected lethal T. gondii infection in murine model of acute toxoplasmosis
AN3661 shows mean IC50 of 32 nM against Plasmodium falciparum laboratory-adapted strains, Ugandan field isolates (mean ex vivo IC50 64 nM), and murine P. berghei and P. falciparum infections (day 4 ED90 0.34 and 0.57 mg kg-1, respectively).
