ACD856 is a potent, pan-Trk positive allosteric modulator (PAM) with EC50 of 382 nM, 295 nM and 33 nM for TrkA, TrkB and TrkC respectively.
ACD856 shows high efficacy on activation of neurotrophin (TrkA, TrkB or TrkC, Emax, 185%, 176%, 328%), with little efficacy on IGF1R and FGFR1.
ACD856 could significantly increase the Vmax(app) of TrkA.
ACD856 (0.1, 0.3 or 1 mg/kg) reverses memory-impairment in the passive avoidance task in mice.
ACD856 improves the acquisition, consolidation, and retrieval of memories.
