A-971432 is a potent, highly selective, CNS-penetrant and orally bioavailable agonist of S1P5 receptor with EC50 values of 4.1 and 5.7 nM in cAMP and GTPγS assays, respectively.
A-971432 displays 60-fold selectivity over S1P1, and >1,600-fold selectivity over S1P2-4, and a panel of 129 kinases and 79 receptors.
A-971432 improves blood–brain barrier integrity in an in vitro model and reverses age-related cognitive decline in mice.
