β-catenin degrader EN83 is a covalent degrader of β-catenin (CTNNB1), depletes CTNNB1 in a ubiquitin-proteasome-dependent manner.
EN83 directly and covalently targets CTNNB1 three cysteines C466, C520, and C619, leading to destabilization and degradation of CTNNB1.
EN83 inhibits CTNNB1 transcriptional activity, demonstrating functional inhibition of the WNT pathway in cells.
EN83 treatment significantly reduced the levels of CTNNB1 target gene MYC.
