eIF4E-RBM38 inhibitor 094 (Compound 094) is a first-in-class small molecule inhibitor of eIF4E-RBM38 complex, binds to eIF4E via its C-terminus pocket (Kd=16 uM) and dissociates RBM38 from eIF4E, enhances p53 translation in RBM38- and eIF4E-dependent manners.
Compound 094 enhances p53 de-novo protein expression in wild-type, but not in eIF4E ∆-Cterm HCT116 cells, enhances p53 expression through enhancing its mRNA translation.
Compound 094 inhibits tumor cell growth and enhances doxorubicin-mediated growth suppression.
