ZYZ384 is a small molecule inhibitor of histone lysine methyltransferase SMYD3 with KD value of 78 uM, reduces cell proliferation against human hepatoma HepG2 cells with IC50 of 5.23 uM.
ZYZ384 inhibits different cancer cell proliferation and suppresses the overexpression of SMYD3.
ZYZ384 (100 mg/kg) promotes growth suppression in HepG2 tumors of nude mice that was better than that of sorafenib at 100 mg/kg.
ZYZ384 reduces the H3K4me3 occupancy at the transcription start site of the Rac1 gene in tumor tissues.
ZYZ384 impairs hepatocellular carcinoma tumor growth in vivo by reducing Rac1 and triggering cell cycle arrest via the AKT pathway.
