ZG297 is a potent, selective S. aureus caseinolytic protease P (SaClpP) agonist with EC50 of 0.26 uM, without activity against Homo sapiens ClpP (HsClpP) (IC50>100 uM), exerts SaClpP-dependent antistaphylococcal activity.
ZG297 directly binds and thermally stabilizes SaClpP protein in S. aureus rather than targeting HsClpP in mammalian cells.
ZG297 exhibits antistaphylococcal activity in a SaClpP-dependent manner.
ZG297 exhibits promising antistaphylococcal activity against S. aureus strains USA300, NRS1, NRS70, NRS108, and NRS271, with low MIC values ranging between 0.063 and 0.125 μg/mL, also exhibits excellent antistaphylococcal activity against MRSA strains, including XJ009, XJ049, XJ051, and XJ052.
ZG297 is a safe and potent antibiotic for treating MRSA infections in vivo.
