ZCZ011 is a positive allosteric modulator of CB1 receptor, increases specific binding of CB1 receptor orthosteric agonist [3H]CP55,940 to mouse brain membranes with pEC50 of 6.9.
ZCZ011 also produced a significant and concentration-dependent increase in the specific binding of the CB1 receptor orthosteric agonist [3H]WIN55212 to mouse brain membranes with pEC50 of 6.3.
ZCZ011 significantly enhanced AEA-stimulated [35S]GTPγS binding at 100 nM.
ZCZ011 potentiated β-arrestin recruitment, and ERK phosphorylation assays.
ZCZ011 is brain penetrant, exhibits antinociceptive effects in neuropathic and inflammatory pain models with no associated cannabimimetic effects.
