YAK540 is a potent, highly selective class IIa HDAC inhibitor with IC50 of 114 nM (HDAC4), displays excellent selectivity against HDAC2 (265-fold), HDAC6 (100-fold), and HDAC8 (82-fold).
YAK540 demonstrates synergistic cytotoxic effect when combined with the proteasome inhibitor bortezomib (BTZ).
The combination of YAK540 and BTZ showed generally increased proapoptotic gene expression, increased p21 expression, and synergistic, caspase 3/7-mediated apoptosis.
