XL20 is a brain-penetrant small molecule targeting the TDP-43 conserved α-helical region spanning residues 320-340 (conserved region, CR) to suppress neurotoxicity, binds to full-length TDP-43 and CR peptide with SPR KD of 10.4 uM and 7.84 uM.
XL20 does not affect TDP-43 splicing activity.
XL20 demonstrates strong neuroprotection at concentrations as low as 6.25 µM in primary cortical neurons overexpressing TDP-43.
XL20 significantly increases the thermal stability of WT TDP-43 in cell lysates.
XL20 selectively suppresses CR-dependent TDP-43 assembly behaviors including LLPS and aggregation, and confers potent neuroprotection at low micromolar concentrations.
XL20 exhibits good CNS penetration.
XL20 attenuates TDP-43 neurotoxicity in mice and human motor neurons, slows disease progression in mutant TDP-43 transgenic mice.
