WB737 is a potent and selective STAT3 inhibitor, directly binds to the STAT3 Src homology 2 (SH2) domain with KD value of 1.34 nM.
WB737 selectively binds to STAT3 over STAT family proteins, including STAT1 and STAT2.
WB737 effectively suppresses cell viability in the three STAT3 Mut NKTL cell lines (SNK6, YT, and NKYS) in a dose‐dependent manner, with limited effects on STAT3 WT cells.
WB737 selectively inhibits the growth of STAT3Mut natural killer/T‐cell lymphoma (NKTL) cells by inducing apoptosis and blocking colony formation.
WB737 selectively inhibits both canonical and noncanonical STAT3 signaling through suppression of pTyr705‐STAT3 and pSer727‐STAT3 in STAT3 Mut cells.
WB737 (5 mg/kg for 14 days, oral gavage) induces almost complete tumor regression in STAT3Mut natural killer/T‐cell lymphoma (NKTL) in YT xenografts mice.
