W2A-28 is a dual modulator of class I HDACs and Wnt/β-catenin signaling, inhibits HDAC1, 2 and 3 with IC50 of 0.51 μM, 0.68 μM and 0.22 μM, respectively, activates Wnt/β-catenin signaling with EC50 of 1.61 μM in Wnt-3A-expressing HEK293 cells.
W2A-28 has no inhibitory effects on other HDACs.
Upregulation of LRP6 protein is associated with W2A-28-mediated activation of Wnt/β-catenin signaling.
W2A-28 induces histone acetylation and activates Wnt/β-catenin signaling in AD patient-specific iPSC-derived cerebral organoids.
W2A-28 significantly inhibits tau phosphorylation in AD patient-specific iPSC-derived cerebral organoids.
