VU6027459 is a potent, selective and CNS-penetrant mGlu7 positive allosteric modulator (PAM) with EC50 of 1.6 uM (rat mGlu7).
VU6027459 is inactive (EC50 > 30 μM, >10 μM at mGlu8) at the other mGlu receptors.
VU6027459 displays CNS penetration in both mice (Kp = 2.74) and rats (Kp= 4.78), and is orally bioavailable in rats (%F = 69.5).
