VTAC-2 is a specific, direct small molecule activator of human PANK3 (Pantothenate kinase 3) with EC50 of 76.5 nM, shows no effect on the catalytic activity of hPANK1 or hPANK2.
VTAC-2 (1 uM) increased hPanK3 activity to >200% of baseline in the absence of acetyl-CoA and maintained activation at approximately 150% in the presence of 10 μM acetyl-CoA.
VTAC-2 (0, 5, and 20 μM) enhances cellular CoA biosynthesis under physiologically-relevant conditions in HepG2 cells.
VTAC-2 (0, 5, or 20 μM, 24 h) restores CoA levels in both engineered and patient-derived PanK2-deficient cells.
VTAC-2 (20 mg/kg, oral, once daily) stimulateed CoA production in a mouse model of Pantothenate kinase-associated neurodegeneration (PKAN), increased brain CoA in PanK2-deficient mice.
