VSW1198 is a potent, selective geranylgeranyl diphosphate synthase (GGDPS) inhibitor with IC50 of 45 nM, VSW1198 is highly specific for GGDPS over other enzymes in the isoprenoid biosynthesis (IBP) pathway.
VSW1198 is a 3:1 mixture of E- and Z-olefin isomers, potently disrupts protein geranylgeranylation in cells at concentrations as low as 50 nM and induces cytotoxic effects at submicromolar concentrations (EC50=0.19 uM, RPMI-8226 cells).
VSW1198 disrupts Rab geranylgeranylation, leading to inhibition of monoclonal protein trafficking, induction of the unfolded protein response pathway (UPR), and apoptosis in multiple myeloma (MM) cells.
