VK-1850 (VK1850) is a small-molecule inhibitor of the DNA binding activity of EBNA1 (Epstein-Barr nuclear antigen1).
VK-1850 selectively inhibits proliferation of EBV-positive cells (C666-1 CC50=7.6 uM) but not similarly growing EBV-negative cells.
VK-1850 potently inhibits EBNA1-dependent OriP DNA replicationm without effect on EBNA1 protein expression in EBV-positive NPC cell line, C666-1.
VK-1850 significantly blocks EBV-positive tumor growth in vivo (10 mg/kg, intraperitoneally, TGI=91.3%), block EBV gene transcription and alter the cellular TGF-β signaling pathway in NPC tumor xenografts.
