URB447 (URB-447) is a peripherally restricted CB(1) antagonist/CB(2) agonist with IC50 of 313 nM (rat CB1) and 41 nM (human CB2).
URB447 inhibited GTPγS binding induced by the CB agonist WIN55,212-2 (1 μM) with EC50 of 4.9 uM in rat cerebellar membrane preparations.
URB447 (1 μM) inhibited cAMP accumulation induced by the β-adrenergic agonist isoproterenol (10 μM) in Hek-293 cells stably expressing mouse CB2 receptors.
URB447 (20 mg/kg, i.p.) drastically reduced total food consumption in male Swiss mice, did not alter motor activity.
URB447 (20 mg/kg, i.p.) selectively prevented peripherally mediated CB responses in mice.
