UCP1162 is a potent propargyl-linked inhibitor of dihydrofolate reductase (DHFR) with IC50 of 21.8 nM for inhibition of DHFR to catalyze the NADPH-dependent reduction of dihydrofolate (DHF) to tetrahydrofolate (THF).
UCP1162 inhibits a panel of cancer cell lines previously reported to be sensitive to antifolates (HCT116, CCRF-CEM, THP-1, and MV-4–11). with iC50 of 0.78-3.3 nM.
UCP1162 also inhibits cancer cell proliferation and bound cellular DHFR at low nanomolar concentrations.
UCP1162 reduced acute inflammation in mice and inhibited FLS cell growth and motility.
