RAD6 SMI#9 (TZ9, RAD6 inhibitor 9) is a specific small molecule inhibitor of ubiquitin-conjugating enzyme (E2) Rad6, inhibits Rad6B-ubiquitin thioester formation and subsequent ubiquitin transfer to histone H2A.

RAD6 SMI#9 inhibited MDA-MB-231 cell proliferation with IC50 of 6 uM.

Rad6 SMI#9 (10 uM) induces MDA-MB-231 G2–M-arrest and decreases β-catenin protein levels, downregulates β-catenin.

Gold nanoparticle (GNP)-based SMI#9 inhibited the TNBC growth in BRCA1 wild-type and BRCA1-mutant TNBC xenograft models.
