TXY436 is an N-Mannich base derivative and FtsZ-directed prodrug of PC190723, TXY436 is effective against S. aureus can also target Gram-negative bacteria, such as Escherichia coli.
TXY436 binds to E. coli FtsZ and inhibit its polymerization/bundling in vitro, exhibits chemical inhibition of AcrAB in E. coli, as well as its homologs in Klebsiella pneumoniae and Acinetobacter baumannii.
TXY436 prodrug exhibits antistaphylococcal efficacy in vivo after oral administration in a murine model of systemic infection.
