TTBK1 inhibitor 62 is a potent, selective, brain-penetrant ATP-competitive Tau tubulin kinase 1 (TTBK1) inhibitor with IC50 of 0.89 uM, >100-fold selective over TTBK2.
TTBK1 inhibitor 62 significantly decreases TDP-43 phosphorylation at Ser409/410 in in SH-SY5Y cells.
TTBK1 inhibitor 62 significantly restores nuclear TDP-43 localization.
TTBK1 inhibitor 62 does not elicit ciliogenesis-related toxicity.
TTBK1 inhibitor 62 restored cognitive deficits, conferred neuroprotection in the frontal cortex, and reduced microglial activation in FTD CaMKIIα-TDP-43 mouse model.
