TTA-A2 is a potent, selective, state-dependent T-type calcium channel antagonist with IC50 of 89 nM and 4100 nM in voltage clamp assays with -80-mV and -100-mV holding membrane potentials in HEK 293 cells heterologously expressing human CaV3.1, respectively.
TTA-A2 displays no significant activities against L-, N-, P/Q-, and R-type calcium channels.
TTA-A2 decreases inactive phase food intake and accentuates diurnal rhythms in core temperature and activity during HFD.
TTA-A2 reduces body weight and improves body composition in diet-induced obese rats.
