TMP-316 (BPN-0037316) is a selective inhibitor of protein kinase A, G and C (AGC) family with cell-free Kd of 1.1 nM and 0.017 nM for S6K1 and ROCK2, shows high selectivity against the broader kinome.
TMP-316 docks into the ATP-binding cleft of all four representatives (ROCK2, S6K1, PKX, PKCγ) with a similar type I binding pose engaging conserved hinge, G-rich, and DFG-in residues.
TMP-316 remodels neuronal signaling to drive neurite outgrowth.
TMP-316 (0.3 or 1 mg/kg) promotes locomotor recovery in a rat cervical hemicontusion model.
