StemRegenin 1 (SR1) is a potent, selective antagonist of aryl hydrocarbon receptor (AHR) with IC50 of 127 nM, promotes the expansion of human hematopoietic stem cell (HSC).
SR1 promotes the retention of CD34 expression following ex vivo culture with an EC50 of 120 nM.
SR1 also expands the number of colony forming cells (CFU).
SR1 only weakly inhibits TCDD-induced transcription in murine cells and has no activity on rat cells, suggesting that SR1 preferentially inhibits human AhR.
SR1 inhibits AhR photoaffinity ligand (PAL) in liver cytosol isolated from hAhR transgenic mice with IC50 of 40 nM in competitive binding assays.
