Simufilam hydrochloride (PTI-125) is a small molecule targeting scaffolding protein filamin A (FLNA), preferentially binds altered FLNA and restores its native conformation.
PTI-125 restores receptor and synaptic activities and reducing its α7nAChR/TLR4 associations and downstream pathologies.
PTI-125 reduces tau hyperphosphorylation, aggregated Aβ42 deposition, neurofibrillary tangles, and neuroinflammation in triple-transgenic (3xTg) Alzheimer's disease (AD) mice.
PTI-125 alleviates neuronal abnormalities and reduced seizure frequency in mice modeling TSC and FCDII with mutations in PI3K-ras homolog enriched in brain (Rheb) pathway genes.
