SW155246 is a potent, selective antagonis/inhibitor of DNA methyltransferase DNMT1 with IC50 of 1.2 uM against hDNMT1, 30-fold preference versus human or murine DNMT3A or -3B (IC50=38 uM).
SW155246 showed a variable degree of cell-growth inhibition against HeLa and A549 with IC50 of 8-20 uM.
SW155246 inhibited global methylation in HeLa cells, reactivated expression of the tumor suppressor gene RASSF1A in A549 cells.
