SP3-12 is a novel selective small molecule inhibitor of Src Homology 2-containing Inositol 5'-Phosphatase-1 (SHIP-1, encoded by INPP5D) with IC50 of 6.1 uM and Ki of 3.2 µM, 7-fold selectivity over SHIP-2.
SP3-12 significantly improved phagocytosis, achieving an EC50 of 2.0 µM in the pHrodo-myelin assay using human microglial clone 3 (HMC3) cells.
SP3-12 represents a novel class of small molecule SHIP-1 inhibitors, demonstrating both selectivity and cellular activity.
