SIRT4-IN-1 (Sirt4 inhibitor 69) is a highly isoform selective inhibitor of mitochondrial deacylase Sirtuin 4 (Sirt4) with IC50 of 16 uM.
SIRT4-IN-1 displays high selectivity against Sirt1, −2, −3, −5, and −6.
SIRT4-IN-1 inhibits Sirt4 through binding to active site regions overlapping with the acyl-peptide binding regions but not with the NAD+ site.
SIRT4-IN-1 dose-dependently increases GDH activity in both whole cells and mitochondrial extract of C2C12 mouse myoblast cells.
SIRT4-IN-1 is active in cells and can serve as a chemical tool for physiological studies on Sirt4.
