生物&药学活性

SIP0401 is a potent, isoform-preferential PDE4B inhibitor with IC50 of 282 nM (PDE-Glo) and 338.4 nM (HTRF), selective for PDE4B over PDE4A/C/D (IC50=1.16-1.97 uM).
SIP0401 induces concentration-dependent cAMP accumulation in THP-1 macrophages with EC50 of 2.52 uM.
SIP0401 dose-dependently inhibits LPS-induced TNF-α secretion in differentiated THP-1 macrophages (IC50=3.24 uM), similar to the reference PDE4 inhibitor rolipram.
