SHP2 inhibitor LY6 (LY6) is a small molecule inhibitor that stabilizes the autoinhibited conformation of the oncogenic tyrosine phosphatase SHP2, inhibits SHP2 with IC50 of 9.8 uM, 7-fold selectivity over SHP.
SHP2 inhibitor LY6 inhibits full length WT SHP2, full length SHP2 E76K, and the SHP2 PTP domain with IC50 of 9.8, 7.67 and20.87 uM, respectively.
SHP2 inhibitor LY6 effectively inhibits SHP2-mediated cell signaling and proliferation, mouse and patient leukemia cells with PTPN11 activating mutations were more sensitive to this inhibitor than wild-type cells.
