生物&药学活性

SH514 is a specific small molecule inhibitor of interferon regulatory factor 4 (IRF4) with IC50 of 2.63 uM, inhibits proliferation of IRF4-high-expressing NCI-H929 and MM.1R MM cells with IC50 of 0.08 uM and 0.11 uM, respectively.
SH514 binds to the IRF4-DBD domain with SPR KD of 1.28 uM.
SH514 shows no effect on the stability of IRF3.
SH514 selectively inhibits proliferation of IRF4-high-expressing cells, regulates IRF4 downstream gene expression, inhibits the protein expression of CDC2, Cyclin B1, Cyclin D1, Cyclin E1, and CMYC.
SH514 (12.5-25 mg/kg once daily) effectively inhibits MM tumor growth in NCI–H929 subcutaneous tumor model.
